Fluorescence Microscopy of Cereals

چکیده

The fluorescence microscope is one of the most sensitive instruments available for morphological and microchemical analysis of biological material, and especially of cereal grains. Recent innovations in illuminating systems, fluorescence chemistry, and specimen preparation have combined to provide significant improvements over conventional bright-field microscopy in both specificity and sensitivity. A variety of relatively specific fluorescent markers has been devised for routine and high resolution detection of all major cereal components. Several examples of useful fluorescent markers are described, including appropriate methods for specimen preparation, fluorescence analysis, and photography. Initial paper received February 25, 1982. Final manuscript received July 16, 1982. Direct inquiries to R.G. Fulcher. Telephone number: 613-995-9428.

A Thin Layer Imaging with the Total Internal Reflection Fluorescence Microscopy

Total internal reflection fluorescence microscopy (TIRFM) is an optical technique that allows imaging of a thin layer of the sample with a thickness of about 100-200 nm. It is used in science of cell biology to study cellular processes, especially near the membranes of living cells. This method is based on the total internal reflection phenomenon, where the evanescent wave is generated in the l...

B-Glucans in the Caryopsis of Sorghum Bicolor (L.) Moench

Fluorescence microscopy was used to determine the l ocation of Bglucans in sorghum. Sections from three genetically different sorghum cultivars were st ained with Calcofluor or Congo Red , fluorochromes which have been reported to react with 13-glucans. Autofluorescence, indicative of ferulic acid in other cereals, was observed in untreated sections. When stained sections were treated with endo...

Determination of 2,4-Dichlorophenoxyacetic Acid in Cereals by Fluorescence Polarization Immunoassay

A fluorescence polarization immunoassay (FPIA) procedure is developed for the determination of 2,4-dichlorophenoxyacetic acid in cereal grains using a Sentry-200 portable device. We synthesized tracers, that is, antigens labeled with f luorescein derivatives, based on two fluorescent compounds and two chlorophenoxyacids. The tracer synthesized from 2,4-dichlorophenoxyacetic acid and 4-aminometh...

Enzymic Mechanism of Starch Breakdown in Germinating Rice Seeds: 8. Immunohistochemical Localization of beta-Amylase.

Rabbit antiserum against beta-amylase isolated from germinating seeds of rice was produced, and its specific cross-reactivity with beta-amylase was confirmed by means of Ouchterlony double immunodiffusion and immunoelectrophoresis procedures. The cellular localization of beta-amylase was studied by indirect fluorescence microscopy of thin sectioned germinating rice seed specimens (1-day stage) ...
